== Isolation, structure determination, and methods of analysis == Steroid isolation, depending on context, is the isolation of chemical matter required for chemical structure elucidation, derivitzation or degradation chemistry, biological testing, and other research needs (generally milligrams to grams, but often more or the isolation of "analytical quantities" of the substance of interest (where the focus is on identifying and quantifying the substance (for example, in biological tissue or fluid). The amount isolated depends on the analytical method, but is generally less than one microgram. The methods of isolation to achieve the two scales of product are distinct, but include extraction, precipitation, adsorption, chromatography, and crystallization. ==== Zinc intoxication ==== Zinc intoxication may cause anemia by blocking the absorption of copper from the stomach and duodenum. Zinc also upregulates the expression of chelator metallothionein in enterocytes, which are the majority of cells in the intestinal epithelium. Since copper has a higher affinity for metallothionein than zinc, the copper will remain bound inside the enterocyte, which will be later eliminated through the lumen. This mechanism is exploited therapeutically to achieve negative balance in Wilson's disease, which involves an excess of copper. But in copper-deficient individuals, zinc excess may cause this mechanism to further deplete copper levels.

=== Synaptic formation and repair === The most-substantiated role for APP is in synaptic formation and repair; its expression is upregulated during neuronal differentiation and after neural injury. Roles in cell signaling, long-term potentiation, and cell adhesion have been proposed and supported by as-yet limited research. In particular, similarities in post-translational processing have invited comparisons to the signaling role of the surface receptor protein Notch. APP knockout mice are viable and have relatively minor phenotypic effects including impaired long-term potentiation and memory loss without general neuron loss. A 2025 mouse study reported that conditional deletion of APP family proteins in excitatory forebrain neurons reduced synaptic localization of the NMDAR subunit GluN1 and suppressed spontaneous cortical and hippocampal neuronal activity, supporting a role for APP family proteins in NMDAR-dependent synaptic and circuit function in vivo. Furthermore, transgenic mice with upregulated APP expression have also been reported to show impaired long-term potentiation. The logical inference is that because Aβ accumulates excessively in Alzheimer's disease its precursor, APP, would be elevated as well. However, neuronal cell bodies contain less APP as a function of their proximity to amyloid plaques. The data indicate that this deficit in APP results from a decline in production rather than an increase in catalysis. Loss of a neuron's APP may affect physiological deficits that contribute to dementia.

=== Cell culture === Connecting the DMF chip to use in the field or world-to-chip interfaces have been accomplished by means of manual pumps and reservoirs which deliver microbes, cells, and media to the device. The lack of extensive pumps and valves allow for elaborate multi step applications involving cells performed in a simple and compact system. In one application, microbial cultures have been transferred onto the chip and allowed to grow with the use of sterile procedures and temperature required for microbial incubation. To validate that this was a viable space for microbial growth, a transformation assay was carried out in the device. This involves exposing E.coli to a vector and heat shocking the bacteria until they take up the DNA. This is then followed by running a DNA gel to assure that the wanted vector was taken up by the bacteria. This study found that the DNA indeed was taken up by the bacteria and expressed as predicted. Human cells have also been manipulated in Digital Microfluidic Immunocytochemistry in Single Cells (DISC) where DMF platforms were used to culture and use antibodies to label phosphorylated proteins in the cell. Cultured cells are then removed and taken off chip for screening. Another technique synthesizes hydrogels within DMF platforms. This process uses electrodes to deliver reagents to produce the hydrogel, and delivery of cell culture reagents for absorption into the gel. The hydrogels are an improvement over 2D cell culture because 3D cell culture have increased cell-cell interactions and cel-extracellular matrix interactions.
==== Phase 1 ==== For the first phase, the WHO formed a team of ten researchers with expertise in virology, public health and animals to conduct a thorough study. One of the team's tasks was to retrospectively ascertain what wildlife was being sold in local wet markets in Wuhan. The WHO's phase one team arrived and quarantined in Wuhan, Hubei, China in January 2021. Members of the team included Thea Fisher, John Watson, Marion Koopmans, Dominic Dwyer, Vladimir Dedkov, Hung Nguyen-Viet, Fabian Leendertz, Peter Daszak, Farag El Moubasher, and Ken Maeda. The team also included five WHO experts led by Peter Ben Embarek, two Food and Agriculture Organization representatives, and two representatives from the World Organisation for Animal Health. The inclusion of Peter Daszak in the team stirred controversy. Daszak is the head of EcoHealth Alliance, a nonprofit that studies spillover events, and has been a longtime collaborator of over 15 years with Shi Zhengli, Wuhan Institute of Virology's director of the Center for Emerging Infectious Diseases. While Daszak is highly knowledgeable about Chinese laboratories and the emergence of diseases in the area, his close connection with the WIV was seen by some as a conflict of interest in the WHO's study. When a BBC News journalist asked about his relationship with the WIV, Daszak said, "We file our papers, it's all there for everyone to see." The team was denied access to raw data, including the list of early patients, swabs, and blood samples. It was allowed only a few hours of supervised access to the Wuhan Institute of Virology.
