Quality control for freeze-dried forms includes visual inspection, water content measurement, and reconstitution time. A satisfactory cake is typically uniform, porous, and intact, although minor shrinkage or cracking may be acceptable if specifications allow. Karl Fischer titration, thermal gravimetric analysis, and near-infrared spectroscopy are used to measure water content. Reconstitution is assessed by adding a specified diluent and recording the time and ease of dissolution. Microbiological and particulate tests are added when the product is sterile or intended for injection.
Stability of a lyophilized solid depends on water content, temperature, and the physical state of the formulation. Amorphous products may slowly absorb moisture and drop below their glass transition temperature, causing collapse or crystallization. Some proteins and peptides can aggregate even in a dry state, especially when exposed to heat or moisture. Accelerated stability studies at elevated temperature and humidity help estimate shelf life, but real-time data remain the basis for expiration dating.
The physical chemistry of freezing influences whether a formulation forms an amorphous glass or a crystalline solid. Amorphous systems can collapse if product temperature rises above the glass transition temperature of the freeze concentrate. Crystalline systems may show eutectic melting, where ice and solute melt together at a fixed temperature. Formulators add bulking agents, lyoprotectants, and buffers to preserve structure and biological activity. The optimum cycle keeps product temperature below critical thresholds during primary drying while allowing efficient sublimation.
Lyophilization is a dehydration process that removes water from a frozen material by sublimation under low pressure. The material is first frozen so that water becomes ice; then the surrounding pressure is lowered below the vapor pressure of ice. Heat is applied gently so ice changes directly to vapor without passing through a bulk liquid phase. The vapor is collected on a cold condenser, leaving a dry porous matrix. This process differs from simple evaporation because the material remains frozen during the main drying stage.
The process usually has three stages: freezing, primary drying, and secondary drying. Freezing sets the ice crystal structure and can determine pore size in the final cake. Primary drying removes free ice by sublimation at low pressure and low shelf temperature. Secondary drying removes bound water by raising the shelf temperature, although some water may remain as part of the solid. Cycle parameters depend on formulation, fill volume, vial type, and equipment performance.
| Property | Value | Notes |
|---|---|---|
| Appearance | Porous cake or plug | Uniform structure suggests the drying cycle preserved the matrix. |
| Reconstitution time | Usually under 2 minutes | Depends on cake porosity, diluent volume, and excipient composition. |
| Water content range | 0.5–3% w/w | Common specification range; exact limits are product-specific. |
| Headspace oxygen | <1% v/v | Inert gas backfill reduces oxidation of sensitive materials. |
| Storage temperature | 2–8 °C or controlled room temperature | Choice depends on accelerated and real-time stability results. |
Stability programs monitor lyophilized products under defined temperature and humidity conditions over time. Real-time studies at recommended storage conditions are the reference, while accelerated studies provide early signals of degradation pathways. Because a dry cake can still undergo oxidation, hydrolysis, or aggregation, stability depends on residual moisture, excipients, and container headspace. Open questions include how best to predict long-term stability from short accelerated runs and how vial-to-vial variability affects shelf life. Current guidance treats these predictions as product-specific rather than universally generalizable.
Freeze-dried materials are hygroscopic to varying degrees and can take up moisture after drying. Storage therefore often uses sealed glass vials, rubber stoppers, and crimp seals to limit contact with ambient humidity. A desiccant may be included for moisture-sensitive products, although it is not universal. Controlled room temperature is sufficient for many lyophilizates, while others require refrigeration or freezing. Moisture ingress remains a primary cause of cake collapse, chemical degradation, and loss of reconstitution performance.
The physics of freeze-drying couples heat transfer, mass transfer, and phase change. Heat supplied through the shelf must reach the sublimation front without melting the ice or degrading the product. Water vapor then travels through the already dried layer and leaves the chamber, where low pressure and cold traps keep it from returning. The dried layer acts as a resistance to vapor flow, so drying rate changes as the front recedes. Open questions remain about how pore structure and formulation heterogeneity affect drying uniformity at larger scales.
Lyophilization removes water from a frozen material by sublimation under reduced pressure. The process begins with freezing, which converts liquid water into ice and concentrates dissolved solids. Primary drying then lowers chamber pressure so ice changes directly into vapor without passing through a liquid phase. Secondary drying raises the shelf temperature to remove bound water that remains after ice sublimation. The result is a dry, porous structure that can be reconstituted later.
Formulation composition influences whether freeze-drying produces an intact cake or a collapsed mass. Excipients such as sugars and polymers can raise the collapse temperature and provide bulk during drying. The critical temperature for primary drying is often the collapse temperature or the glass transition temperature of the maximally concentrated phase. If the product temperature exceeds this threshold, the frozen matrix may soften and lose structure. Established practice therefore links shelf temperature and chamber pressure to the formulation's thermal properties.
== Antiviral drug resistance == Antiviral resistance can be defined by a decreased susceptibility to a drug caused by changes in viral genotypes. In cases of antiviral resistance, drugs have either diminished or no effectiveness against their target virus. The issue inevitably remains a major obstacle to antiviral therapy as it has developed to almost all specific and effective antimicrobials, including antiviral agents. The Centers for Disease Control and Prevention (CDC) inclusively recommends anyone six months and older to get a yearly vaccination to protect them from influenza A viruses (H1N1) and (H3N2) and up to two influenza B viruses (depending on the vaccination). Comprehensive protection starts by ensuring vaccinations are current and complete. However, vaccines are preventative and are not generally used once a patient has been infected with a virus. Additionally, the availability of these vaccines can be limited based on financial or locational reasons which can prevent the effectiveness of herd immunity, making effective antivirals a necessity. The three FDA-approved neuraminidase antiviral flu drugs available in the United States, recommended by the CDC, include: oseltamivir (Tamiflu), zanamivir (Relenza), and peramivir (Rapivab). Influenza antiviral resistance often results from changes occurring in neuraminidase and hemagglutinin proteins on the viral surface. Currently, neuraminidase inhibitors (NAIs) are the most frequently prescribed antivirals because they are effective against both influenza A and B.
===== Hop effectors ===== HopZ1s are type III effectors which interfere with the Glycine max 2-hydroxyisoflavanone dehydratase (GmHID1). HopZ1b degrades daidzein after production, reducing concentrations and thus reducing the immunity it provides the plant.
=== 2024–present: I'm a Celebrity... South Africa and Everything. All At Once === In April 2024, Collins made a guest appearance on the E4 reality series The Underdog: Josh Must Win, in which contestants believe they are competing on a reality show called The Favourite, however unbeknownst to them, the game is being controlled by Amber Gill, Nick Grimshaw, Pete Wicks and Vicky Pattison who must ensure that Josh, a non-typical reality show contestant, is declared the winner. Collins was enlisted to help with a photoshoot in which she assigned costumes to the contestants and advised them on various different poses. In May 2024, Collins appeared on the Daily Mail's podcast series Everything I Know About Me where she spoke about her life and career. In September 2024, Collins appeared on the BBC genealogy documentary series Who Do You Think You Are?. Collins was keen to learn more about her mother's side of the family, after she was fostered at birth and ultimately discovered that her grandmother was a diagnosed chronic schizophrenic who spent time in two psychiatric hospitals. Throughout the episode, she meets her distant cousins who reside in Dagenham near to where Collins grew up, and sees a photo of her grandmother for the first time. She also discovers that her ancestors lived on Dorset Street in Spitalfields, where Jack the Ripper is believed to have murdered his final victim Mary Jane Kelly in November 1888. In January 2025, she appeared on an episode of Pointless Celebrities alongside Hayley Hasselhoff, filmed two years prior.
=== Hazards === When taken in prescribed quantities, codeine-promethazine syrup is quite safe, but dangers arise in higher doses since it can cause one to stop breathing. Using alcohol and other drugs alongside lean increases the chance of serious adverse effects. The drink does not appear to cause seizures itself, but can increase their likelihood in those already susceptible to them. It has been suggested that the promethazine in the drink may heighten the euphoric effects of the codeine. The addictive nature of the drink means that trying to discontinue regular usage can bring about symptoms of opioid withdrawal. In a 2008 interview with MTV News, Lil Wayne described the withdrawal as feeling "like death in your stomach when you stop. Everybody wants me to stop all this and all that. It ain't that easy."
== Absorption, metabolism and excretion == In the upper small intestine, thiamine phosphate esters present in food are hydrolyzed by alkaline phosphatase enzymes. At low concentrations (<2 μmol l−1), the absorption process is carrier-mediated. At higher concentrations, absorption also occurs via passive diffusion. Active transport can be inhibited by alcohol consumption or by folate deficiency. The majority of thiamine in serum is circulating bound to albumin, with over (90%) in erythrocytes (red blood cells), and is delivered to cells with high metabolic needs—particularly those in the brain, liver, pancreas, heart, and skeletal and smooth muscles, including cardiac muscle cells. A specific binding protein called thiamine-binding protein has been identified in rat serum and is believed to be a hormone-regulated carrier protein important for tissue distribution of thiamine. Uptake of thiamine by cells of the blood and other tissues occurs via active transport and passive diffusion. Two members of the family of transporter proteins encoded by the genes SLC19A2 and SLC19A3 are capable of thiamine transport. In some tissues, thiamine uptake and secretion appear to be mediated by a Na+-dependent transporter and a transcellular proton gradient. Human storage of thiamine is about 25 to 50 mg, with the greatest concentrations in liver, skeletal muscle, heart, brain, and kidneys. ThMP and free (unphosphorylated) thiamine are present in plasma, milk, cerebrospinal fluid, and, it is presumed, all extracellular fluid.

Anti-Peruvian (Spanish: Antiperuanismo), or Peruvophobia, refers to negative feelings, fear, hatred and discrimination toward and/or against Peruvians based on a combination of historical, cultural, and ethnic prejudices. It arose since the 19th century in some societies as a consequence of their territorial expansion. It germinated as a tendency in the nationalisms of neighboring countries, mainly Ecuador, Chile and to a lesser extent due to the disputed origin of different cultural manifestations, such as recipes and gastronomic preparations (such as pisco or picarones) or folkloric dances (such as the diablada or the morenada) whose origin is disputed or shared with Chile and Bolivia. In addition, due to different political and ideological differences with the Bolivarian leaders and their Chavista sympathizers in Venezuela. It can manifest itself in many ways, such as individual hatred or discrimination, tabloid media, attacks by groups organized for that purpose, even on social networks.
== Factors/pathways == Cells can detect and react to mechanical stimuli in a variety of ways. One method is through the interaction of E-cadherin presented on the cell membrane. As these receptors interact and are pulled or pushed, tension can be created, leading to a change in the conformation of alpha-catenin bound to B-catenin on the intracellular portion of E-cadherin. This causes the recruitment of vinculin and leads to a change in actin conformation and in the orientation of the cell. Another signaling pathway important in a cell's response to mechanical stimuli is the Wnt planar cell polarity (PCP) pathway. This noncanonical pathway involves the activation of Rho and Rac families of GTPases, which are essential in reorganizing the cytoskeleton in preparation for cell migration. When cells collide, localized signaling of the PCP pathway leads to a change in the polarity of the cell, redirecting the cell in a different direction. Different cellular receptors are important in cellular mechanotransduction involved in contact with a substrate such as the extracellular matrix (ECM). For example, many cell types express a5b1 integrin on their membranes, which can bind to a major ECM component called fibronectin. This leads to an accumulation of integrins in the area of contact with the ECM, attaching the ECM to the cytoskeleton of the cell and allowing for migration to occur along the ECM through tension at the points of attachment (called focal adhesions, FA) and subsequently the dismantling of FAs as the cell moves along.
The company had originally been incorporated as Tim Donut Limited. By the 1990s, the company name had changed to The TDL Group Ltd. This was an effort by the company to diversify the business, removing the primary emphasis on donuts, and continuing the expansion of the menu options as consumer tastes broadened. Some older locations retain signage with the company's name, including a possessive apostrophe, despite the fact that the official styling of the company's name has been Tim Hortons without an apostrophe for at least a decade. The company had removed the apostrophe after signs using the apostrophe was interpreted by some to be breaking the language sign laws of the province of Quebec in 1993. The removal of the apostrophe allowed the company to have one common sign image across Canada. Although a number of Quebec locations have bilingual menu boards, the decision to have both Canadian official languages represented is left to the discretion of individual franchise owners. Some Quebec locations have French-only menu boards. It is the strong recommendation to all the Quebec restaurants from the TDL Group Corporation that they post menu boards in both English and French in accordance with the standards being enforced by the Office québécois de la langue française.
On January 3, 2026, the United States launched airstrikes in Venezuela, capturing Venezuelan president Nicolás Maduro and his wife, and resulting in an estimated 80 casualties, including Maduro's personal guard, military personnel, and civilians. The airstrike was done without congressional awareness or authorization, resulting in criticism from media and a number of congressmen. Maduro and his wife were arraigned in a Manhattan federal court on 5, January 2026. Both pleaded not guilty to a number of drug trafficking charges. Followed by a purge of Maduro-era loyalists from the Venezuelan government, which was taken over by Delcy Rodríguez. Since 2026, Venezuela has been a de facto puppet state of the United States. The Trump administration, primarily through U.S. Secretary of State Marco Rubio, maintains control over its domestic finances, foreign policy, government appointments, revenues, and oil reserves.
=== Allergy === An allergic reaction, formally known as type 1 hypersensitivity, is the result of an inappropriate immune response triggering inflammation, vasodilation, and nerve irritation. A common example is hay fever, which is caused by a hypersensitive response by mast cells to allergens. Pre-sensitised mast cells respond by degranulating, releasing vasoactive chemicals such as histamine. These chemicals propagate an excessive inflammatory response characterised by blood vessel dilation, production of pro-inflammatory molecules, cytokine release, and recruitment of leukocytes. Severe inflammatory response may mature into a systemic response known as anaphylaxis.

Sealed vials or containers should be kept at the temperature specified by stability data, often controlled room temperature or 2–8 °C. Moisture and oxygen barriers are important because both can degrade sensitive materials. Opened containers may need immediate use or protection from ambient humidity.
It usually appears as a uniform porous plug or cake that fills the container without excessive shrinkage. Color should match the specification, and there should be no meltback or visible foreign matter. Minor cracking may be acceptable if the product still meets moisture and potency limits.
Water content is a key stability parameter because excess water can promote hydrolysis, aggregation, or cake collapse. It also affects reconstitution and product weight. Each product has a target range, and methods such as Karl Fischer titration are used to verify it.
The main change is sublimation, in which ice becomes water vapor without melting into liquid water. This occurs when the chamber pressure is held below the vapor pressure of ice while mild heat is supplied. The result is a dry, porous solid that retains much of its original shape.
