The process usually has three stages: freezing, primary drying, and secondary drying. Freezing sets the ice crystal structure and can determine pore size in the final cake. Primary drying removes free ice by sublimation at low pressure and low shelf temperature. Secondary drying removes bound water by raising the shelf temperature, although some water may remain as part of the solid. Cycle parameters depend on formulation, fill volume, vial type, and equipment performance.
The physical chemistry of freezing influences whether a formulation forms an amorphous glass or a crystalline solid. Amorphous systems can collapse if product temperature rises above the glass transition temperature of the freeze concentrate. Crystalline systems may show eutectic melting, where ice and solute melt together at a fixed temperature. Formulators add bulking agents, lyoprotectants, and buffers to preserve structure and biological activity. The optimum cycle keeps product temperature below critical thresholds during primary drying while allowing efficient sublimation.
Lyophilization is a dehydration process that removes water from a frozen material by sublimation under low pressure. The material is first frozen so that water becomes ice; then the surrounding pressure is lowered below the vapor pressure of ice. Heat is applied gently so ice changes directly to vapor without passing through a bulk liquid phase. The vapor is collected on a cold condenser, leaving a dry porous matrix. This process differs from simple evaporation because the material remains frozen during the main drying stage.
Stability programs monitor lyophilized products under defined temperature and humidity conditions over time. Real-time studies at recommended storage conditions are the reference, while accelerated studies provide early signals of degradation pathways. Because a dry cake can still undergo oxidation, hydrolysis, or aggregation, stability depends on residual moisture, excipients, and container headspace. Open questions include how best to predict long-term stability from short accelerated runs and how vial-to-vial variability affects shelf life. Current guidance treats these predictions as product-specific rather than universally generalizable.
Freeze-dried materials are hygroscopic to varying degrees and can take up moisture after drying. Storage therefore often uses sealed glass vials, rubber stoppers, and crimp seals to limit contact with ambient humidity. A desiccant may be included for moisture-sensitive products, although it is not universal. Controlled room temperature is sufficient for many lyophilizates, while others require refrigeration or freezing. Moisture ingress remains a primary cause of cake collapse, chemical degradation, and loss of reconstitution performance.

Formulation composition influences whether freeze-drying produces an intact cake or a collapsed mass. Excipients such as sugars and polymers can raise the collapse temperature and provide bulk during drying. The critical temperature for primary drying is often the collapse temperature or the glass transition temperature of the maximally concentrated phase. If the product temperature exceeds this threshold, the frozen matrix may soften and lose structure. Established practice therefore links shelf temperature and chamber pressure to the formulation's thermal properties.
The physics of freeze-drying couples heat transfer, mass transfer, and phase change. Heat supplied through the shelf must reach the sublimation front without melting the ice or degrading the product. Water vapor then travels through the already dried layer and leaves the chamber, where low pressure and cold traps keep it from returning. The dried layer acts as a resistance to vapor flow, so drying rate changes as the front recedes. Open questions remain about how pore structure and formulation heterogeneity affect drying uniformity at larger scales.
Quality control for lyophilized products focuses on appearance, moisture level, reconstitution time, and structural integrity. A cake should be uniform, intact, and free of meltback or collapse. Moisture level is measured by Karl Fischer titration or thermogravimetric analysis. Reconstitution time reflects pore structure and formulation. Visual inspection and vial integrity checks detect cracks, stopper defects, or particulate matter. These checks are often performed on samples from each batch. They help confirm that the drying cycle performed as intended.
Storage stability depends on water content, oxygen exposure, and temperature. Lyophilized solids are hygroscopic and can absorb water if exposed to humid air. Vials are usually sealed under vacuum or inert gas with rubber stoppers and aluminum crimps. Storage temperatures range from room temperature to refrigerated or frozen conditions depending on the material. Stability studies track potency, moisture, and physical form over time. Accelerated conditions can reveal sensitivity but may not predict long-term behavior.
Freezing determines ice crystal structure and pore size, which affect drying speed and product uniformity. Rapid freezing creates small crystals, while slow freezing creates larger crystals and often faster sublimation. During primary drying, chamber pressure is held below the vapor pressure of ice, and shelf temperature supplies heat for sublimation. The ice front recedes, leaving a porous matrix. Thermal limits such as collapse and eutectic temperatures set safe boundaries for formulation. These limits vary with solute composition and concentration.
Secondary drying removes bound water that remains after ice sublimation. Shelf temperature is raised gradually while pressure remains low, reducing water content to a target range. Over-drying can cause brittleness or electrostatic issues, while under-drying affects stability. The endpoint is often judged by pressure rise tests, temperature measurements, or water content analysis. Scale-up depends on matching heat and mass transfer across equipment sizes. Small changes in shelf temperature or pressure can alter cycle length substantially.
Lyophilization, or freeze-drying, removes water from a material by freezing it and then lowering pressure so ice changes directly to vapor. The process relies on sublimation, the phase transition from solid to gas without an intervening liquid state. It is used for heat-sensitive materials that would degrade in conventional drying. The three stages are freezing, primary drying, and secondary drying, each with distinct temperature and pressure requirements. In practice, cycle design balances these variables.
Reconstitution involves adding a suitable diluent, often sterile water or a buffer, to the dried cake. Gentle swirling or inversion helps dissolve the material without creating excessive foam. The time required for complete dissolution can range from seconds to several minutes and depends on the cake structure and the diluent. Improper reconstitution, such as vigorous shaking or using the wrong diluent, can cause protein aggregation or loss of activity. After reconstitution, the product may have a limited shelf life and should be used according to its labeling.
Lyophilized products are typically hygroscopic and require protection from moisture during storage. Manufacturers seal them in glass vials, often under vacuum or an inert gas such as nitrogen. The container closure system, including the stopper and crimp seal, must prevent water vapor ingress. Storage temperature varies from controlled room temperature to refrigerated or frozen conditions, depending on the formulation. Humidity-controlled environments are essential because even brief exposure to ambient air can degrade the product.
Stability of a lyophilized product depends on its glass transition temperature, the temperature at which the amorphous cake transitions from a glassy to a rubbery state. Storage below this temperature minimizes molecular mobility and slows chemical degradation. If the storage temperature exceeds the glass transition temperature, the cake may collapse, shrink, or become sticky. Accelerated stability studies at elevated temperatures and humidity help predict shelf life, but they do not always reflect real-time behavior. Residual moisture content also plays a critical role in long-term stability.
== History == Sauk City restaurateurs and husband and wife team George and Ruth Culver started their fast food careers as the owners of an A&W on Phillips Boulevard (U.S. Highway 12) in 1961. In 1968, they purchased a resort-styled restaurant at Devil's Lake called The Farm Kitchen. Their son, Craig Culver, worked for a local McDonald's right out of college in 1973. == In culture == Black peppercorns were found stuffed in the nostrils of Ramesses II, placed there as part of the mummification rituals shortly after his death in 1213 BCE. Little else is known about the use of pepper in ancient Egypt and how it reached the Nile from the Malabar Coast of India.
== Flow control == There are various ways to control the fluid flow in the channels. They include changing the channel width and length, altering the wettability of the paper, diverting some fluid through a parallel channel, or changing the viscosity of the fluid. The flow in PADs can be turned off with dissolvable sugar bridges, Corona discharge treatment to alter a coating on the paper from a hydrophobic to hydrophilic state, or the use of an expandable polymer triggered by the flow to close the flow path.
=== Ferdinand Schwarzwald === Ferdinand Schwarzwald (Nico Rogner) is the CFO of family-owned Austrian bank IBN Bauer, and a director on the board of Tender, in which his bank has a 6% stake. He backs Whitney's vision for pivoting the company away from pornography and gambling clients and into banking, and cautions Whitney that his co-founder and CEO Jonah Atterbury is "not a serious person", contributing to Jonah's eventual removal from the company. Ferdinand later helps Whitney and new Tender CEO Henry Muck execute a merger with IBN Bauer, traveling with them to Vienna to assuage the concerns of family heir Moritz-Hunter Bauer. He and Whitney also procure a billion-dollar investment from Al-Mi'raj Pierpoint during the release of Tender's banking app. Ferdinand is later revealed to be Whitney's handler on behalf of Russian foreign intelligence, having been recruited years ago by Cozy Bear (the hacker group affiliated with the FSB and SVR) and installed as IBN Bauer's CFO after stealing client lists that Russia could use as leverage. Amid public revelations of Tender's fraudulent profits out of Africa, Ferdinand warns Whitney that their narrative must remain intact, and implies that the Russians can assassinate anyone who goes "off-message". Ferdinand and fixer Dez Watkins later catch Whitney trying to disappear via New York and intercept him, forcing him to proceed with a hostile takeover bid for Pierpoint as it would represent a valuable "data set" for Russia.
Bukele passed a law in 2021 that made bitcoin legal tender in El Salvador and promoted plans to build Bitcoin City. In June 2023, the Legislative Assembly approved Bukele's proposals to reduce the number of municipalities from 262 to 44 and the number of seats in the legislature from 84 to 60. He ran for re-election in the 2024 presidential election and won with 85 percent of the vote after the Supreme Court of Justice reinterpreted the constitution's ban on consecutive re-election. Bukele is highly popular in El Salvador and throughout Latin America. He holds an average approval rating of 88.6% during his entire presidency with approval ratings never having fallen below 75%. El Salvador has experienced democratic backsliding under Bukele's leadership, falling 61 places in the World Press Freedom Index between 2019 and 2025 and 24 places in the Economist Intelligence Unit's Democracy Index, which now classifies El Salvador as a hybrid regime. In February 2020, Bukele ordered 40 soldiers into the Legislative Assembly building to intimidate lawmakers into approving a US$109 million loan for the Territorial Control Plan, an event that triggered a political crisis and was described by the opposition as a self-coup. After Nuevas Ideas won a supermajority in the 2021 legislative election, Bukele's allies in the legislature voted to replace the attorney general and all five justices of the Supreme Court of Justice's Constitutional Chamber. Bukele has criticized journalists and news outlets, and has furthered press censorship.

This is a list of radioactive nuclides (sometimes also called isotopes), ordered by half-life from shortest to longest, in seconds, minutes, hours, days and years. Current methods make it difficult to measure half-lives between approximately 10−19 and 10−10 seconds.
Strigolactones (SLs) were originally discovered through studies of the germination of the parasitic weed Striga lutea. It was found that the germination of Striga species was stimulated by the presence of a compound exuded by the roots of its host plant. It was later shown that SLs that are exuded into the soil also promote the growth of symbiotic arbuscular mycorrhizal (AM) fungi. More recently, another role of SLs was identified in the inhibition of shoot branching. This discovery of the role of SLs in shoot branching led to a dramatic increase in the interest in these hormones, and it has since been shown that SLs play important roles in leaf senescence, phosphate starvation response, salt tolerance, and light signalling.
=== Hypothalamus === Temperature is regulated in the hypothalamus. The trigger of a fever, called a pyrogen, results in the release of prostaglandin E2 (PGE2). PGE2 in turn acts on the hypothalamus, which creates a systemic response in the body, causing heat-generating effects to match a new higher temperature set point. There are four receptors in which PGE2 can bind (EP1-4), with a previous study showing the EP3 subtype is what mediates the fever response. Hence, the hypothalamus can be seen as working like a thermostat. When the set point is raised, the body increases its temperature through both active generation of heat and retention of heat. Peripheral vasoconstriction both reduces heat loss through the skin and causes the person to feel cold. Norepinephrine increases thermogenesis in brown adipose tissue, and muscle contraction through shivering raises the metabolic rate. If these measures are insufficient to make the blood temperature in the brain match the new set point in the hypothalamus, the brain orchestrates heat effector mechanisms via the autonomic nervous system or primary motor center for shivering. retinal + NAD+ + H2O → retinoic acid + NADH + H+ Retinoic acid is actively transported into the cell nucleus by CRABp2 where it regulates thousands of genes by binding directly to gene targets via retinoic acid receptors. In addition to retinol, retinal and retinoic acid, there are plant-, fungi- or bacteria-sourced carotenoids which can be metabolized to retinol, and are thus vitamin A vitamers. There are also what are referred to as 2nd, 3rd and 4th generation retinoids which are not considered vitamin A vitamers because they cannot be converted to retinol, retinal or all-trans-retinoic acid. Some are prescription drugs, oral or topical, for various indications. Examples are etretinate, acitretin, adapalene, bexarotene, tazarotene and trifarotene.

The main change is sublimation, in which ice becomes water vapor without melting into liquid water. This occurs when the chamber pressure is held below the vapor pressure of ice while mild heat is supplied. The result is a dry, porous solid that retains much of its original shape.
Freezing determines ice crystal size, solute distribution, and the pore network left after drying. A slow or fast freezing rate can produce different cake structures and affect reconstitution. It also sets whether the formulation follows an amorphous or crystalline drying path.
It removes most free water during primary drying and part of the bound water during secondary drying. A small residual moisture content often remains and is specified for each product. Complete removal is generally neither practical nor desirable for stability.
Many dried cakes are hygroscopic and can adsorb water during storage or handling. Absorbed moisture may lower the glass transition temperature and promote chemical reactions. Sealed packaging and controlled humidity reduce this risk.
